Loss of ERb expression as a common step in estrogen-dependent tumor progression

چکیده
The characterization of estrogen receptor beta (ERb) brought new insight into the mechanisms underlying estrogen signaling. Estrogen induction of cell proliferation is a crucial step in carcinogenesis of gynecologic target tissues, and the mitogenic effects of estrogen in these tissues (such as breast, endometrium and ovary) are well documented both in vitro and in vivo. There is also an emerging body of evidence that colon and prostate cancer growth is influenced by estrogens. In all of these tissues, most studies have shown decreased ERb expression in cancer as compared with benign tumors or normal tissues, whereas ERa expression persists. The loss of ERb expression in cancer cells could reflect tumor cell dedifferentiation but may also represent a critical stage in estrogen-dependent tumor progression. Modulation of the expression of ERa target genes by ERb or ERb-specific gene induction could explain that ERb has a differential effect on proliferation as compared with ERa. ERb may exert a protective effect and thus constitute a new target for hormone therapy, such as ligand specific activation. The potential distinct roles of ERa and ERb expression in carcinogenesis, as suggested by experimental and clinical data, are discussed in this review. Endocrine-Related Cancer (2004) 11 537–551

متن کاملEstrogen Receptors A and B Are Inhibitory Modifiers of Apc- Dependent Tumorigenesis in the Proximal Colon of Min/+ Mice
Estrogen replacement therapy in postmenopausal women is associated with a reduction in colorectal cancer risk, potentially via interactions between 17B-estradiol (E2) and the estrogen receptors (ER) A and B. To study the role of E2 in intestinal tumor inhibition, we separately crossed C57BL/ 6J-Min/+ (Min/+) mice with Era and Erb mice to generate ER-deficient Min/+ progeny. We found an increase...
متن کاملتأثیر هورمون استروژن بر میزان پروتئین p53 در رده سلولی T47D سرطان پستان
Background: Breast cancer is one of the most common cancers in women. Nearly 30% of breast cancers are hormone-dependent, and these hormones comprising estrogens influence progression of breast cancers. It is now widely recognized that p53 may be the most frequently mutated protein in breast cancer. High levels of p53 protein are a common feature of many human malignant cancers. Given that, T47...
متن کاملPreclinical Development Therapeutic Significance of Estrogen Receptor b Agonists in Gliomas
Gliomas are the most common and devastating central nervous system neoplasms. A gender bias exists in their development: females are at lower risk than males, implicating estrogen-mediated protective effects. Estrogen functions are mediated by two estrogen receptor (ER) subtypes: ERa, which functions as tumor promoter, and ERb, which functions as tumor suppressor. We examined the potential use ...

Graphical Abstract Highlights d Prostate tumorigenesis caused by PTEN deletion involves loss of estrogen receptor b d ERb transcription is repressed by BMI-1, which is induced by PTEN deletion d ERb repression is needed for tumorigenesis because it enables HIF/VEGF signaling d HIF/VEGF signaling sustains BMI-1 expression, resulting in a positive feedback loop A causal role for ERb in prostate c...
